CFU Processing Using Digital Imaging & Differential Staining [NYBC592]
An objective, standardized high throughput semi-automated system for the enumeration and characterization of Colony Forming Units (CFU) which represent the hematopoietic potential of umbilical cord blood stem cells to be used for hematopoietic reconstitution via transplantation. The system utilizes high resolution digital imaging (HRDI) that captures an image of the culture dish containing the Colony Forming Units (CFU) and provides a means to easily identify different colony types. The system uses built-in positive sample identification and storage of the image along with assay results for future reference. This system overcomes the current lack of standardization of methods and variability of results encountered in the characterization and enumeration of CFUs performed under light microscopy. Further, current methods do not allow for re-counting or visualization of a sample after the assay is complete, as sample plates are discarded at the end of the 14 day assay.
